Study on the Synthesis of Acetanilide under Microwave Radiation

The acetanilide was synthesized under the action of microwave irradiation from phenylamine,glacial acetic acid that acted as raw material and zinc that acted as catalyst.The factors that influence the yield were studied,and the optimized reaction conditions were determined,i.e.reaction temperature was 100℃,microwave irradiation power was(600 W) irradiation time was(15 min),and the ratio of phenylamine to glacial acetic was 1∶2.5.And under this reaction conditions,the yield of acetanilide was enhanced from 55% to 70%,reaction time was shortened,and the reaction rate was raised.

here are three major methods to purify products. The first is Distillation, which only can be utilized when the product is liquid phase. When using the Distillation method, TLC (Thin Layer Chromatography) is able to use to monitor the reaction going. It is really easy to tract on the process by doing this way. Recrystallization is the second technique, and it is used to purify solids with a small amound of impurities. Unforturnitely, Thin Layer Chromatography can not be utilized when using this method. The third one is Colomn Chromatography. It can be used to purify any mixture of solid and liquid with the difference of polarity.Thin Layer Chromatography is utilized to monitor the separating process, to diffirenciate the good separation and poor separation. This assay is focucing only on the Second method, Recrystallization.

Synthesis of Acetanilide was started from aniline and acetic anhydride via a nucleophilic acyl substitution reaction to form acetanilide and acetid acid under water (base).Under a basic condition, the free electrons in amine(aniline) attracts the C2 of Acetic anhydride (carbonyl group of acyl compound) and formed tetrahedral intermediate. The leaving group acetate was reppled by reforming of keton. The free electon on H2O attract the H in NH2+ cation and formed H3O+,and left product acetanilide. Also the acetate pulled a proton from H3O+ to form acetid acid, while the H3O+ was going back to be H2O.

Choosing of solvent

The most difficult techinique in recrystallization is picking a suitable recrystallization solvent. The best solvent shuld be in an ideal system, which means the compound should be completely soluble in the hot solvent and quite insoluble in cold solvent, while the impurities should be either soluble at all temperature or insoluble at all temperature. Through experiments, distilled water would be the best solvent for this recrystallization. H2O is polar compound. the polarity is similar to acetanilide, but not the identical. It is the other condition to satify the 'best solvent'

Starting of Recrystallization

The hot solvent, water, should be as minimum as possible to dissolve the crude product. After all the product has been dissolved, the solution should be cooled slowly to form the crystals. Changing temperature rapidly would result the preciptating, rather then recrystallizing. Cooling temperature slowly gives the solution time to form the solid lattice. The specific solid lattice would not allow the impurites to involve, therefore that's the purification.

All the crystals obtained from the lab should go through drying procedure. Vaccum separators are recommented, because of high efficiency of separating solid and liquid through funnels.

Example experimental of synthesis Acetanilide

Aniline(2 ml,2.06 g,22.1 mmol), Acetic anhydride(3 ml,3.24 g,31.7 mmol), and distilled water(15 ml,15 g, 0.832 mol) were combined and stirred 30 minutes. Upon completion, the product was dried by porcelain Buchner funnel and ice-cold water(2 ml) was used to wash product for 20 minutes to dry off solvent. Remaining white power was desired product. The melting point of pure acetanilide should be the range from 113 C to 115 C.
